While sperm cells are made in the testicles, the actual seminal fluid that carries the sperm is made in the seminal vesicles, which are directly behind the prostate. The vas deferens carries sperm from the testes up into the seminal ducts where it mixes with the seminal fluid as it collects in the prostatic urethra to create semen. This mixture builds up during sexual activity and is temporarily held in the prostatic urethra until orgasm. While blue balls is a mismomer, the pain is caused by semen building up in a very narrow space. Finally, slightly alkaline prostatic fluid is released during the first contractions of orgasm, and is one of the reasons semen tastes bad.
So back to the original point, sperm is created in the testes, but the balls do not hold any semen in them. In fact, ejaculation is even possible without testes.
